Master's Thesis from the year 1998 in the subject Biology - Genetics / Gene Technology, grade: 11 on 13-scale, , language: Danish, abstract: Dette project handlede om en sekvens p? 22,62 kb, som indeholder 17 nyligt sekventeret gener i Bacillus subtilis, h.h.v. yut M, yut K, yut J, yut I, yut H, yut G, yut F, yut E, yut D, yut C, yunA, yun B, yunC, yunC, yunE, yunF, og yunG. Form?let med projektet var f?rst at mutagenisere disse ukendte gener, som skulle dern?st analyseres og sammenlignes med vildtypen. Mutationerne blev udf?rt ved at indf?re plasmidet pMUTIN4 efter en standardiserede metode udviklet for B. Subtilis Genomprojektet. Der blev konstrueret over 56 stammer med defekter i 15 af de aktuelle gener. Stammerne indeholdte lacZ i starten af generne, hvilket muliggjorde at unders?ge, hvorn?r generne blev udtrykt, ved blandt andet udpladning p? LB-plader med og uden IPTG. Mutant stammerne blev dern?st ved en southern blot analyse unders?gt for, om de havde en eller flere kopier af plasmidet inkorporeret p? kromosomalt DNA. Southern blottet indikerede, at inkorporeringen af plasmidet var som ?nsket (en kopi) i 14 mutant stammer, imens der var en mutant stamme af yutJ med tandem insert. Derudover de gentagne mislykkede transformations fors?g (10 fors?g) for yunA og yunG understregede at disse to gener var essentielle for overlevelse. Homologis?gningen viste, at nogle af de 17 gener var st?digv?k ukendte s?som yutD, yutC, yunB, yunC,yunF og yunG, hvorimod nolge af dem havde analogi med nogle allerede karakteriserede gener enten fra B. subtil eller fra andre organismer. F?notypisk ekspressions unders?gelser viste, at ligesom yutE, yutD og yutF, generne yunB, yunC, yunD, yunE, yunF, og yunG var organiseret i en operon. Biographie:
Widad Akrawi is a geneticist and global health expert, and known for being a Danish human rights defender, peace advocate, and author. She was born 1969 in Akre, Kurdistan region. She graduated as a civil engineer in 1990, and proceeded on to the Technical University of Denmark where she earned a Masters degree in genetics and microbiology. After graduation, she served as a clinical geneticist and researcher in human genetics and inherited diseases at the Danish Royal Hospital, the largest hospital in Denmark and a leading university hospital in Northern Europe. She has also earned a PhD degree in global health and epidemiology.In 2003, she became a published writer. After researching the concept of victim psychology and the effects of various types of trauma on individuals' psychological health, she published her first novel, Tara's Book, written in Danish, in Copenhagen. The book was translated to Norwegian in 2005, published in Oslo, and distributed by the Norwegian Cultural Council to almost all Norwegian libraries. Her eBook, entitled Under H?bets Skyggee (Under the Shadow of Hope), also in Danish, was published in 2013 at Amazon.com and Lulu.com. She currently leads Defend International, a Norway-based NGO that is devoted to medical research, and human rights. Over the years, she has addressed and actively participated in several regional and international meetings and conferences, such as the United Nations Third and Fourth Biennial Meeting of States, the UN Commission on The Status of Women, the General Assembly Interactive Hearings with Non-Governmental organizations, Civil society organizations and the Private sector, and the UN First Committee on Disarmament and International Security. She has held numerous leadership positions in national and international organizations, including executive board member of Amnesty International Denmark, ambassador for Amnesty International's Stop Torture campaign, and member of the Panel of Vestby Experts on approaches to socio-cultural constructivism. At present, she is a board member of Women of Europe Award, an active member of International Action Network on Small Arms (IANSA), a board member of IANSA Women's Network Working Group, and a senior lobbyist at the UN, lobbing for the adoption of an international Arms Trade Treaty during the negotiations that take place at the UN General Assembly's First Committee on Disarmament and International Security.
